Il padre, Adam Anastos Belushi era un immigrato albanese che abbandono la cittadina dove era cresciuto, Qyteze (era nato a Coriza) nel 1934 all'eta di sedici anni e dopo 21 anni a Chicago apri due ristoranti (era partito da semplice commesso). In Albania la famiglia si chiamava Bellios, ma il cognome cambio in Belushi col trasferimento negli Stati Uniti. La madre, Agnes Demetri Samaras (nata nel 1922) era una farmacista americana nata ad Akron, nell'Ohio, dove si era trasferita la sua famiglia nel 1915 anch'essa dall'Albania (il padre era nato a Korysta nel 1864 e la madre lo stesso anno in Grecia). John aveva una sorella, Marian, piu grande di 12 anni e due fratelli piu piccoli: James Adam Belushi o "Jim" (piu piccolo di 5 anni), e William Adam Belushi o "Billy," nato il giorno dell'undicesimo compleanno di John (24 gennaio 1960).
